Coming from Charlotte, the I-81 route is best and keeps you on
interstate most of the way.

Rt 301 is (my opinion) horrible because of all the stop lights.  But
then I lived in the DC area enough that I have no problems with
it's traffic.  But either way, the 301 route is definitely not for
you because it presupposes you're going through Richmond
and I
